Oncolytic viral therapy is a treatment that relies on tumor specific replication of viruses leading to their lytic destruction. The lecture will cover genetic manipulations which give HSV-1 tumor specificity. This will include examples of vectors in clinical trials currently being investigated for safety and efficacy in GBM patients. The lecture will also discuss preclinical research that has uncovered changes in the tumor microenvironment upon OV therapy, and the significance of these responses in modulating anti-tumor efficacy. Some approaches to modify OV therapy to improve therapeutic response will also be discussed.
